It's hard to just pick one best desperation or peeing scene.

I like the peeing scene from the Korean movie Call Boy, they made it so dramatic lol.

If I had to pick cartoons, then 6teen and that surfer cartoon made by the same people have good pee scenes, for some reasons the team who made those shows really like body jokes like peeing and farting but surprisingly there's a lot of scenes with girls peeing, like in 6teen she gets frightened and pees in the toilet, another time they have a meeting in the girl's room but she has to go so badly she just uses the next door stall and everyone is listening and shocked. 

For other films I agree that Mary Must Run one is really amazing. There was a similar (Dutch?) film that students shot called Harndrang?, it's very similar to Mary Must Run so you should check it out, has a woman basically try to use public toilets but the lines are too long and she can't find a public toilet anywhere.

Buffalo '66 all the way. It's one of my favorite movies, and re-watching it last June made me realize I had this fetish and it completely changed my life. The entire opening of the film is about 15 minutes of the main character looking for a bathroom after being released from prison. There isn't a lot of physical desperation, but he gets increasingly frustrated. After a few hours, he caves and pisses on the sidewalk. Great movie, great showcase of desperation (MALE desperation, which is rare enough as it is).

Is it the best? I dunno, that's subjective, but it's what got me into the show and highly attracted to this character~
